A synthetic pentadecapeptide derived from a fragment of human gastric juice protein, studied extensively in rodent models for gastrointestinal, tendon, ligament, and muscle healing.
Overview
BPC-157 is a synthetic pentadecapeptide (15 amino acids) derived from a naturally occurring protective protein found in human gastric juice. Since the 1990s, a research group led by Predrag Sikiric at the University of Zagreb has published the large majority of the animal literature on BPC-157, examining its effects on gastrointestinal healing, tendon and ligament repair, and muscle-to-bone reattachment in rat models.
Proposed Mechanism of Action
Across the cited studies, BPC-157's effects are attributed primarily to pro-angiogenic activity (upregulation of the VEGFR2 pathway) and modulation of the nitric oxide system, both of which are implicated in accelerated tissue repair in the animal models studied. It has not been evaluated in adequately powered human clinical trials.
Research Context
The three studies referenced here span three distinct rodent injury models: a chemically induced gastric ulcer model, a surgically transected medial collateral ligament model, and a myotendinous junction detachment model. All three report accelerated functional and histological recovery in BPC-157–treated animals relative to controls, using both injectable (intraperitoneal, intramuscular) and oral routes.
Limitations of the Current Evidence
- The literature base is dominated by a small number of overlapping research groups, primarily using rat models — this narrows the diversity of experimental replication.
- No large-scale human clinical trial data exists to establish safety, efficacy, or dosing in humans.
- Reported effective doses span several orders of magnitude (picograms to micrograms per kilogram) across studies, indicating the dose-response relationship is not well characterized.
